Supporting data for “DECIPHERING THE OPTIMAL SMALL MOLECULE ACTIVATORS OF HIF-1α STABILIZATION IN DENTAL STEM CELLS”

<p dir="ltr">RNA sequencing of PHD2-knocked-down-SHED was compared to a drug's unique signature, creating a connectivity map. TW-37 emerged as the best compound based on connectivity score and <i>P</i>-value, potentially mimicking the PHD2-knocked-down-SHED transcriptomic signature. The expression of HIF-1α in SHED was detected by western blot under different concentrations, and the vascular endothelial growth factor (VEGF) level was assessed. Live/Dead staining assessed the impact on SHED proliferation and viability. Differential gene expression of TW-37-treated-SHED compared to control-SHED was performed by RNA sequencing, and hub genes were identified to explore the mechanism of TW-37 on HIF-1α activation. To evaluate the effect on SHED' angiogenic properties, we conducted an <i>in-vitro</i> sprouting assay was conducted.</p>
