This program aim to study the role and regulation of gamma-secretase in testis. We first time identify that TNFα is an upstream regulator of gamma-secretase mediated JAM-A cleavage in testis. This cytokine can upregulate the protein level of PS1, PEN2 and MMP-9 via p38 MAPK signaling pathway, and thus lead to an intracellular cleavage of JAM-A by gamma-secretase and a extracelluar shedding of JAM-A by MMP-9. Intracellualr fragment after cleavage of JAM-A by gamma-secretase downregulates the protein level of Abcc1, thus may further regulate the restructuring of BTB.
