This is supporting data for my thesis entitled "ARIP4 resolves R loops at promoter proximal regions of highly transcribed, androgen-dependent genes to promote PolII pause release and transcriptional induction".
The promoter proximal region, which is defined as the region in proximity to the promoter where PolII transcription machinery pauses, awaiting further signals for pause release. Recently, this region has been highlighted as a hotspot where cotranscriptional R loops form and DSBs form. However, the regulation of cotranscriptionl R loops, PolII pausing and release, DSB formation, and how this impacts transcriptional output has not been linked together. In this study, we demonstrate that ARIP4, a putative RNA helicase, is a novel R loop helicase. ARIP4 is enriched at the promoter proximal regions of paused genes together with PolII, where it facilitates transcriptional induction by resolving pausing-associated R loops and promote pause release.
This supporting data contains raw data in addition to processed data illustrated in the thesis. The raw data consists of quantification files, uncropped Western Blots and Native PAGE gels, and timelapse photos compiled into a video format of live imaging samples.
