Supporting dataset for "Enhancing remyelination therapy with use of the bone marrow neurosphere-derived pericytes"

The dataset contained all the data for the thesis "Enhancing remyelination therapy with use of the bone marrow neurosphere-derived pericytes" This study showed a protocol to derive brain-pericyte like cells with the potential to treat demyelination from a readily accessible source, bone marrow. With the protocol modified from our lab, rat bone marrow stromal cells were used to induce bone marrow neurospheres in ultra-low adhesion conditions with a neurosphere induction medium. Afterwards, rat bone marrow neurospheres were later seeded on an adherent culture plate coated with gelatin and cultured under a pericyte induction medium to give rise to pericytes.


The supporting data provided the data to show the identity of the bone marrow neurosphere-derived pericytes, suggesting their subtypes and their functions. It contained the characterization of rat and human bone marrow stromal cells, neurospheres, and then the bone marrow neurosphere-derived pericytes to show their identities and high purity in each derivation stage. Furthermore, the dataset provided the statistics of the tube formation assay for rat bone marrow neurosphere-derived pericytes and also the immunocytochemistry data to show the assembly of pericytes in an endothelial network.


Furthermore, the dataset includes the behavioural data to show the effect of rat bone marrow neurosphere-derived pericytes on in vivo remyelination using shiverer model with co-transplantation with rat bone marrow neurosphere-derived oligodendrocyte precursor cells and rat bone marrow neurosphere-derived pericytes. The rotarod performance test measured the functional recovery of motor performance, and co-transplantation of rat bone marrow neurosphere-derived pericytes and rat bone marrow neurosphere-derived oligodendrocyte precursor cells was shown to improve the motor performance of the shiverer mice after 12 weeks of co-transplantation. It also includes the data from immunohistochemistry to show the presence of the transplanted cells after one week of transplantation, showing the survival of the transplanted cells.
