Metabolic dysfunction-associated steatotic liver disease (MASLD) is a leading form of chronic liver disease due to rising incidences in obesity and insulin resistance. We first screened 6 metabolites on HepG2 cells to evaluate its effect on lipid metabolism by qPCR and RNA-sequencing. We found that 2-aminoadipic acid (AAA) and alpha aminobutyric acid (ABA) -treated cells showed enrichment in steroid biosynthesis and lipid metabolism. Subsequently, we administered AAA and ABA in a diet-induced MASLD mice model and investigated its effect on hepatic triglyceride accumulation, lipid metabolism, cholesterol metabolism, and bile acid metabolism.
